Screening of five essential oils for identification of potential inhibitors
Publicaçãopor Neves, AOutros Autores: Rosa, S; Gonçalves, J; Rufino, A; Judas, F; Salgueiro, L et al.Nuclear factor-kappaB is a key transcription factor activated by pro-inflammatory signals, like interleukin-1beta (IL-1), being required for the expression of many inflammatory and catabolic mediators, such as nitric oxide (NO), that play an important role in arthritic diseases. This work aimed at screening and identifying natural inhibitors of IL-induced NF-kappaB activation and NO production in human articular chondrocytes. Five essential oils obtained from four plants of the Iberian flora, Mentha x piperita L. (Lamiaceae), Origanum virens L. (Lamiaceae), Lavandula luiseri L. (Lamiaceae), and Juniperus oxycedrus L. subsp. oxycedrus (Cupressaceae), were screened for their ability to prevent IL-1-induced NO production. The oil showing higher inhibitory activity was fractionated, concentrated, analyzed for composition elucidation and prepared for further assays. For this purpose, the human chondrocytic cell line C-28/I2 was used to evaluate NF-kappaB activation by determining the cytoplasmic levels of the total and phosphorylated forms of the inhibitory protein, I kappaB-alpha, and the NF-kappaB-DNA binding activity. The essential oil from the leaves of J. oxycedrus in a concentration of 0.02 % (v/v) achieved the greatest inhibition (80 +/- 8%) of IL-1-induced NO production. Chemical analysis showed that this essential oil is predominantly composed of monoterpene hydrocabons, being alpha-pinene [2,6,6-trimethyl-bicyclo(3.1.1)hept-3-ene] the major constituent (76 %). Similarly to the effect of the whole oil, a fraction containing 93% alpha-pinene reduced significantly IL-1-induced I kappaB-alpha degradation. Moreover, alpha-pinene also decreased I kappaB-alpha phosphorylation, NF-kappaB-DNA binding activity, and NO production. Another fraction containing oxygenated mono- and sesquiterpenes was nearly as effective as alpha-pinene. The ability of the alpha-pinene-containing fraction to reduce IL-1-induced NF-kappaB activation and NO production warrants further studies to demonstrate the usefulness of alpha-pinene in the treatment of arthritic diseases and other conditions in which NF-kappaB and NO play pathological roles. -

Negative impact on the reproductive potential of blue jack mackerel Trachurus picturatus by Kudoa infection of the ovary
Publicaçãopor Neves, A.Outros Autores: Gordo, I.; Sequeira, V.; Vieira, A.R.; Silva, E.; Silva, F. et al.Origem: IC-onlineReproduction of Trachurus picturatus off the western Portuguese coast was studied over 1 yr. During histological analyses, the presence of Kudoa sp. was detected in advanced vitellogenic oocytes. Kudoa infections are known to cause economic loss through the induction of postmortem liquefaction of fish muscles, but ovarian infection as reported in this study will seriously affect the reproductive potential of the species and thus impact fisheries productivity. Only females showed gonad infection which led to total degradation of advanced vitellogenic oocytes. No macroscopic, somatic or condition indices revealed differences between infected and uninfected females, rendering this infection event a concealed suppression of reproduction. -

Reproduction of the blue jack mackerel, Trachurus picturatus, in western Portugal: microscopic gonad analysis reveals indeterminate fecundity and skipped spawning patterns
Publicaçãopor Neves A.Outros Autores: Sequeira V.; Vieira A.R.; Silva E.; Silva F., Duarte A.M.; Mendes S. et al.Blue jack mackerel, Trachurus picturatus, is the fifth most landed fish species in mainland Portugal, but information on its reproductive biology is scarce. From September 2018 to August 2019, 626 specimens were collected from commercial vessels to clarify the reproductive strategy of the T. picturatus population off the west coast of Portugal. The proportion and length range of males and females were similar. Only three of the specimens collected were categorized as immature, indicating that the fish caught in the fishery are primarily mature. The spawning season lasted from late January until the end of March, with gonadosomatic indices being similar for males and females. Fecundity was indeterminate, and estimated batch fecundity ranged between 6,798 (at 25.4 cm TL) and 302,358 oocytes (at 33.8 cm TL). The low number of females showing direct evidence of imminent or recent spawning suggests a low number of spawning events. In addition, 12.7% of females were considered non-reproductive due to ovary abnormalities including parasitic infection by Kudoa species, atretic structures and skipped spawning events. This study highlights the importance of accounting for skipped spawning events and ovary abnormalities in the management of species fisheries. -

Ear Mites: Tackling a Persistent Problem
Publicaçãopor Neves, A.Outros Autores: Mesquita, J.; Vala, Helena; Nóbrega, C.A several number of mites have been associated with ear problems, namely otitis externa (Scott et al., 2001). Fifty percent or more of all otitis, in cats, and 10%, in dogs, are thought to be caused by ear mites (Sotiraki et al., 2001; Mueller, 2008). Very common in pets, ear mites can be a serious problem. The most common mite is, undoubtedly, Otodectes cynotis, but there are some other mites that can also be found in the ear canal, includding Demodex canis, Demodex cati, Sarcoptes scabiei, Notoedres cati, among others (Cordero del Campillo et al., 1999; Blot et al., 2003; Engler, 2007). Frequently, ear mites can be the initiating factor of the otitis externa. The mechanical irritation, caused by their presence inside the ears, may lead to a higher activity of ceruminal glands and, consequently, the establishment of a favorable environment for secondary infections, caused by bacteria or fungi (August (1988) cited by Souza et al. (2008). These secondary infections and the animal’s individual response may perpetuate and induce the progression of the initial disease (Scott et al., 2001). It was established that the number of mites necessary to promote disease can be very low (only about two to three) triggering an Arthus-type and/or an immediate-type hypersensitivity reaction (Scott et al., 2001; Sotiraki et al., 2001; Angus, 2004). It has been reported that young animals tend to be more susceptible to ear mites than adult ones (Blot et al., 2003; Mueller, 2007; Mueller, 2008; Lefkaditis et al., 2009). -

Transparent conductive graphene textile fibers
Publicaçãopor Neves, A. I. S.Outros Autores: Bointon, T. H.; Melo, L. V.; Russo, S.; de Schrijver, I.; Craciun, M. F. et al.Transparent and flexible electrodes are widely used on a variety of substrates such as plastics and glass. Yet, to date, transparent electrodes on a textile substrate have not been explored. The exceptional electrical, mechanical and optical properties of monolayer graphene make it highly attractive as a transparent electrode for applications in wearable electronics. Here, we report the transfer of monolayer graphene, grown by chemical vapor deposition on copper foil, to fibers commonly used by the textile industry. The graphene-coated fibers have a sheet resistance as low as -1 k Omega per square, an equivalent value to the one obtained by the same transfer process onto a Si substrate, with a reduction of only 2.3 per cent in optical transparency while keeping high stability under mechanical stress. With this approach, we successfully achieved the first example of a textile electrode, flexible and truly embedded in a yarn. -
