Catalytic behavior of alkali-treated Pt/HMOR in n-hexane hydroisomerization
Publicaçãopor Monteiro, R.Outros Autores: Ania, C. O.; Rocha, J.; Carvalho, A. P.; Martins, A.Bifunctional Pt-HMOR catalysts were prepared by incipient wetness impregnation of various desilicated MOR obtained by alkaline treatment using NaOH concentrations ranging from 0.1 to 0.5 M. The zeolite structural changes upon modification were investigated by several techniques including powder X-ray diffraction,Al-27 and Si-29 MAS-NMR spectroscopy, N-2 adsorption, pyridine adsorption followed by infrared spectroscopy and the catalytic model reaction of m-xylene transformation. For low alkaline concentration the zeolite acidity is preserved, along with a slight increase of the volume correspondent to the larger micropores due to the removal of extra-framework debris already existent at the parent zeolite. At higher NaOH concentrations there is a significant loss of crystalinity and acidity as well as the formation of mesoporosity. The characterization of the metal function shows similar patterns for Pt-HMOR and Pt-M/0.1 samples, with Pt particles located mainly inside the inner porosity. In contrast, large Pt particles become visible at the intercrystalline mesoporosity of MOR crystals developed during the desilication treatments at severe alkaline conditions. The catalytic results obtained for n-hexane hydroisomerization showed an improved selectivity for dibranched over monobranched isomers for Pt-M/0.1 sample, likely due to the preservation of the support acidity and the slight enlargement of the micropores. This work is a new example in which the mesoporous development does not improve the catalytic efficiency of the zeolites, whereas mild alkaline desilication might be considered as an effective solution to produce customized catalysts with enhanced performance for a given application.

Exploring glutathione as an adjuvant of anti-biofilm strategies against Pseudomonas aeruginosa
Publicaçãopor Monteiro, R.Outros Autores: Pereira, Maria Olívia; Sousa, Ana MargaridaPseudomonas aeruginosa biofilms are involved in several infectious diseases and their eradication remains a challenge. Disrupting the biofilm matrix is the most attractive way to facilitate biofilm-cells eradication and glutathione has been exhibiting great potential as a biofilm disrupter agent. Therefore, this study aimed to explore antibiofilm strategies based on the combination of glutathione with two in-use antibiotics. Biofilms were formed and subjected to the action of combinatorial arrangements of glutathione with ciprofloxacin or tobramycin. Preliminary results showed that PAO1 biofilms were eradicated using glutathione+ciprofloxacin, in contrast to biofilms formed by a clinical isolate (PAI). Better outcomes were obtained using glutathione+tobramycin since biomass reduction occurred with lower dosages. Also, a different action than the usually described for glutathione is suggested. In conclusion, glutathione-tobramycin could be a potential anti-biofilm strategy. -

Deciphering the role of Staphylococcus aureus in Pseudomonas aeruginosa adaptation to cystic fibrosis lungs: bystander or intervener?
Publicaçãopor Monteiro, R.Outros Autores: Magalhães, Andreia P.; Pereira, Maria Olívia; Sousa, Ana MargaridaBackground: Pseudomonas aeruginosa is the major responsible for the high mortality rate of cystic fibrosis (CF) patients due to the development of chronic infections. To long persist, P. aeruginosa uses sophisticated mechanisms to achieve fulladaptation, mainly triggered by the harsh environmental conditions of CF lungs. The influence of Staphylococcus aureus, a primary colonizer of CF lungs, in increasing P. aeruginosa pathogenicity has also been reported. The main objective of this study was to decipher the impact of S. aureus on P. aeruginosa pathogenicity using in vitro conditions mimicking CF lungs. [...] -

Long-term coexistence of Pseudomonas aeruginosa and Staphylococcus aureus using an in vitro cystic fibrosis model
Publicaçãopor Monteiro, R.Outros Autores: Magalhães, Andreia Patrícia Alves; Pereira, Maria Olívia; Sousa, Ana MargaridaAim: To investigate the role of pre-established Staphylococcus aureus on Pseudomonas aeruginosa adaptation and antibiotic tolerance. Materials & methods: Bacteria were cultured mimicking the sequential pattern of lung colonization and exposure to ciprofloxacin. Results: In the absence of ciprofloxacin exposure, S. aureus and P. aeruginosa coexisted supported by the physicochemical characteristics of the artificial sputum medium. S. aureus had no role in P. aeruginosa tolerance against ciprofloxacin and did not select P. aeruginosa small-colony variants during antibiotic treatment. rhlR and psqE were downregulated after the contact with S. aureus indicating that P. aeruginosa attenuated its virulence potential. Conclusion: P. aeruginosa and S. aureus can cohabit in cystic fibrosis airway environment for long-term without significant impact on P. aeruginosa adaptation and antibiotic tolerance. -

Red wine increases adipose tissue aromatase expression and regulates body weight and adipocyte size
Publicaçãopor monteiro, rOutros Autores: soares, r; guerreiro, s; pestana, d; calhau, c; azevedo, iObjective: Obesity is an important component of the metabolic syndrome in constituting a risk factor for cardiovascular disease, diabetes, and cancer. Estrogens influence lipidaccumulation in adipocytes, acting indirectly or directly on adipose tissue. In this study we aimed to investigate the influence of red wine ingestion on the expression of aromatase (estrogen synthase) in adipose tissue. Methods: Red wine or ethanol solution, in the concentration found in red wine, was provided to Wistar rats as the sole drinking fluid for 8 wk. Food and drink intakes and body weight were monitored throughout treatment and adipocyte size and aromatase expression in the adipose tissue were determined at the end of the experimental period. Results: Red wine and ethanol increased aromatase expression in the adipose tissue and red wine decreased adipocyte size (P < 0.05). In addition, animals treated with red wine or ethanol had significantly lower weight gain than controls, despite a similar energy intake. Conclusion: Thus, the ingestion of red wine may alter the production of estrogens by adipose tissue, body weight gain, and adipocyte size. Some of these red wine effects are attributable to ethanol. This relation among estrogen availability, adipocyte biology, and weight gain is most interesting and deserves further study because it may lead to new strategies to reduce metabolic syndrome incidence. Migration and escapement of silver eel males, Anguilla anguilla, from a southwestern European river
Publicaçãopor Monteiro, R.Outros Autores: Domingos, I.; Almeida, P.R.; Costa, J.L.; Alexandre, C.M.; Quintella, B.R.The catadromous life cycle of the European eel encompasses a spawning migration to the Sargasso Sea. Prior to their migration, eels develop a silver coloration being referred to as “silver eels.” Due to the dramatic European eel recruitment decline, it is crucial to quantify silver eel escapement to evaluate the success of measures taken under Eel Management Plans (Regulation EC (1100/2007)). Thus, the percentage of silver eels escaping from the River Mondego (Portugal) was estimated during two consecutive spawning seasons (2014–2016) and their migratory behaviour was studied. Thirty-six male silver eels were tagged with acoustic transmitters and their downstream migration was tracked using automatic receivers deployed at four locations along the river course, from the first impassable obstacle up to the river mouth (67 km). Among the tagged eels, 15 (42%) escaped to the sea. In both years, individuals displayed faster migration speed in the downstream reaches of the river. BRT (“Boosted Regression Trees”) models were used to explain downstream migration and escapement of silver eels. The analyses revealed that the downstream migration is triggered by several environmental variables with water conductivity as the most influential predictor. Escapement of eels from the estuary occurred a few hours after their arrival and was primarily explained by the offshore wind intensity, which promotes a current in the coast that may favour their transport to the open sea. These findings contribute to a better understanding of silver eel migration in southern latitudes and highlight and reinforce the need to monitor spawner escapement.
