Unintentional injuries and associated factors among adolescents
Publicationby Bordalo,DianaOther Authors: Fonseca,Paula; Lopes,Tânia; Rolim,Sara; Figueirinha,Joana; Almeida,Filipa et al.Origin: SciELO PortugalAbstract Introduction: Unintentional injuries are an important cause of death in adolescents, as well as a major cause of ill health. This age group is prone to unintentional injuries due to their specific biological, psychological, and social characteristics. To prevent these injuries, it is crucial to understand their prevalence and associated factors. The aim of this study was to identify and characterize unintentional injuries in adolescents and their potential impact on health and life. Material and Methods: Prospective, observational, two-center study over one year based on a survey of adolescents admitted to the Emergency Department due to unintentional injuries. Results: The study included 1054 adolescents (840 from Center A and 214 from Center B), mostly male (58.2%), 51.6% of which in early adolescence (10 to 13 years old) and only 11.2% in late adolescence (17 to 18 years old). Alchool or drug use were reported by 2.3% of participants. Main reported injuries included traumatic injuries (mainly during sports), accidental falls (mainly at school), and road traffic accidents (being run over by a car as the most frequent). Acidental falls and traumatic injuries were more common in early adolescence, whereas burns, road traffic accidents, and intoxications were more frequent in middle adolescence. Low frequency of personal protection equipment use (31.3%) was reported by bycicle riders. School absence was reported in 31% of cases, mainly associated with traumatic injuries (49.8%) and falls (40.1%). Conclusion: Most reported unintentional injuries had preventable causes and their prevalence was developmental stage-dependent. These results provide additional data for implementation of preventive measures according to developmental adolescence stages. -

Fate map of zebrafish left-right organizer
Publicationby Bordalo, Diana MartinsOrigin: Repositório Institucional da UNLThe organizer is a ciliated signalling transient organ, responsible for the patterning of embryo tissues during embryonic development. In higher vertebrates, such as mouse and chick, this organizer (the node and the Hensen’s node, respectively) performs dorsalventral and anteriorposterior axis definition, as well as left-right patterning of the internal organs. In lower vertebrates, such as frog and zebrafish, there is a separate specialized organ for left-right purposes called the Gastrocoel Roof Plate (GRP) and Kupffer’s Vesicle (KV), respectively. It is known that mouse and chick organizer cells give rise to structures like floor plate, notochord, hypochord and somites. Frog GRP originates all these but floor plate. In zebrafish, at 13-14 somite stage (ss) the KV finished its left-right patterning but what happens to this organizer’ cells is still poorly studied. This research attempts to understand the fate and behaviour of the KV cells. We followed the fate of KV cells by live imaging and by tight time-courses with fixed larvae. We assessed in detail their proliferative and death profile, as well as cilia length progression from 9-10 ss until 29-30 ss. We conclude that the KV cells mostly follow the evolutionarily conserved fates described for other organizers. These cells mainly incorporate the notochord and hypochord; few cells incorporate the floor plate and the somites. As a novelty, it is also hypothesized that the hypural cell fate may be among the KV cell fates. -

Effects of UV filters in Mytilus galloprovincialis sperms and adults under climate change
Publicationby Bordalo, Diana Vanessa MartinsMultiple stressors are leading to unprecedent pressures in coastal systems namely pollutants such as personal care products and climate change such as ocean warming. Avobenzone (AVO) is one of the most used ultraviolet (UV) filters in personal care products which use is progressively higher. UV filters are persistent and can be accumulated in the environment whereas their impacts in marine organisms, in particular bivalves, are poorly studied. Bivalves represent a significant proportion of aquaculture production, being highly abundant in European coastal zones and having socioeconomic importance. Mussels are considered sentinel organisms and are often used in ecotoxicological research. Due to recreational activities at the beach, UV filters reach higher peaks during summer in aquatic bodies, sometimes coincident with the spawning period of mature sedentary mussels. For this reason, this study aims to evaluate the toxic effects of a range of AVO concentrations (0.1, 1.0, 10.0 μg/L) and investigate in which way warming (21 °C) may change the effects caused at different biological levels of the Mediterranean mussel species Mytilus galloprovincialis (Lamarck, 1819): sperm (in vitro 30 min-exposure) and adults (in vivo 28 days-exposure). Effects were assessed in sperm cells (oxidative status, viability, genotoxicity, motility and kinetics) and adults (energy reserves, metabolic capacity, biological defence mechanisms, cell damages and neurotoxicity). Cellular damages occurred only in sperm exposed to warming and their susceptibility to warming increased significantly when combined with AVO, since this UV filter alone did not exert any effects. AVO concentration range, warming and AVO with higher temperature led to oxidative stress in sperm. A decrease of viability was observed in sperm exposed to the highest AVO concentrations and warming. All AVO concentrations induced genotoxicity and warming increased sperm sensitivity to AVO. Respiration rate increased significantly in sperm exposed to 0.5 μg/L AVO, warming and AVO with warming. AVO concentration range and AVO with higher temperature induced a decrease of motility and the highest AVO concentration caused a complete immobility. A decrease of metabolism was observed at the lowest AVO concentration and an increase occurred with warming in adults. Energy reserves and antioxidant defences’ activity increased significantly in adults at the highest AVO concentration. Results suggest a higher sensitivity of sperm regarding AVO and warming, a higher susceptibility to the interactive effect of both stressors and to the highest AVO concentration, with possible impairment of male reproductive functions and disruption of M. galloprovincialis populations. This study highlights the importance of further research on the toxic impacts resulting from the interaction of emergent pollutants and climate change towards bivalves, considering their ecological and economic relevance. Pollution levels in coastal systems tend to increase and mussels’ responses can change under different scenarios, compromising the maintenance of the population, reinforcing the need for more research. -

Dermatology clinical case
Publicationby Vilela Oliveira, RaquelOther Authors: Bordalo, Diana; Carvalho, Sónia; Fonseca, PaulaA previously healthy 15-year-old girl was sent to the Pediatric Emergency Department by the general practitioner due to suspicion of a hematological condition. The girl presented with ecchymotic lesions on the anterior region of the thorax and antero-proximal region of the upper limbs, which had apparently worsened in the last 24 hours. She denied trauma or impact sports, fever or recent infections, and blood loss or other symptoms. Laboratory evaluation showed no alterations and urine toxicology screening was negative. When confronted with normal laboratory evaluation, the girl confessed intentional self-inflicted injuries following a discussion with her mother. Parents were made aware of the situation, and a pediatric psychiatry consultation was recommended. After discharge, follow-up in the girl’s home country was assured. With this report, the author intend to raise awareness of the importance of maintaining a high index of suspicion to non-suicidal self-injury, identifying alarm signs and establishing early preventive strategies. -

Leigh syndrome: a case report with a mitochondrial DNA mutation
Publicationby Lopes, TâniaOther Authors: Coelho, Margarida; Bordalo, Diana; Bandeira, António; Bandeira, Anabela; Vilarinho, Laura et al.Objective: Leigh syndrome is a neurodegenerative disorder with an incidence of 1:40,000 live births. It presents wide clinical, biochemical, and genetic heterogeneity, but with homogenous neuropatoradiological alterations. There is no specific treatment, and the prognosis is reserved. This case report aimed familiarize health professionals with the disease. Case Description: A 16-month-hold girl who was followed in outpatient clinic due to axial hypotonia and delayed psychomotor development. Karyotype, auditory evoked potentials and ophthalmologic evaluation were normal. Evidence of hyperlactacidemia and hypocitrullinemia was detected in the patient. After performing brain magnetic resonance under anesthesia, hypotonia got worse, and the patient was hospitalized after an episode of cyanosis and apnea. The electroencephalogram showed no epileptiform activity. Neuroimaging revealed bilateral lenticular hyperintensity, especially in the putamen and in the left globus pallidus regions. Molecular analysis revealed an 8993T>G (MT-ATP6) mutation in the mitochondrial DNA. Comments: Between 10 and 30% of individuals with Leigh syndrome have mitochondrial DNA mutations. The decompensation after anesthetic intercurrences is typically associated with neurological deterioration and, in this case, increased the diagnosis suspicion. It is important to alert for similar cases and to reduce invasive diagnostic tests if the diagnosis is suspected. -

Salt pan brine water sulphated polysaccharides retrieved at pilot scale: ability to stimulate in vitro human macrophages and salmon head kidney cells
Publicationby Ferreira, Sónia S.Other Authors: Pereira, Renato B.; Bordalo, Diana; Barbosa, Helena; Ferreira, Nicole; Correia, Alexandra et al.Marine environments are the warehouse of a variety of novel bioactive compounds prone to be explored by food and feed industry. The growing interest in sulphated polysaccharides has led to the search for new sustainable sources, such as seawater. These compounds are naturally concentrated in salt pan brine water due to their evaporation by wind and sunlight. To take advantage of these sources, sulphated polysaccharides were concentrated from salt pan brine water using a scalable membrane ultrafiltration system with 30 and 100 kDa cut-off. This process allowed to concentrate ten times the polymeric material of brine water into 1.9 g/L, rendering a fluffy polysaccharide rich material after drying. It was mainly composed of 23 % (w/w) of uronic acids, 19 % of sulphate esters, and 34 % (w/w) of neutral sugars. This polymeric material has shown to stimulate in vitro both human macrophages and Atlantic salmon head kidney SHK-1 cells in a range of 6.25–50 μg/mL without toxicity, showing potential to be used in both human food and aquaculture feeding.
