Isolation, Identification, and Characterization of Phosphate-Solubilizing Bacteria from Tunisian Soils
Publicaçãopor Amri, MarwaOutros Autores: Rjeibi, Mohamed Ridha; Gatrouni, Marwa; Mateus, D. M. R.; Asses, Nedra; Pinho, Henrique J. O. et al.Origem: Instituto Politécnico de TomarSoil microorganisms play an important role in maintaining natural ecological balance through active participation in carbon, nitrogen, sulfur, and phosphorous cycles. Phosphate-solubilizing bacteria (PSB) are of high importance in the rhizosphere, enhancing the solubilization of inorganic phosphorus complexes into soluble forms available for plant nutrition. The investigation of this species of bacteria is of major interest in agriculture, as they can be used as biofertilizers for crops. In the present study, 28 isolates of PSB were obtained after the phosphate enrichment of soil samples from five Tunisian regions. Five PSB species were identified by 16S rRNA gene sequencing including Pseudomonas fluorescens, P. putida, and P. taiwanensis, Stenotrophomonas maltophilia, and Pantoea agglomerans. Solid and liquid Pikovskaya’s (PVK) and National Botanical Research Institute’s (NBRIP) media containing insoluble tricalcium phosphate were used for the evaluation of the phosphate solubilization ability of the bacterial isolates by two methods: visual evaluation of the solubilization zone around colonies (halo) and determination of solubilized phosphates in liquid medium by the colorimetric method of the vanado-molybdate yellow. Based on the results of the halo method, the isolate of each species that showed the higher phosphate solubilization index was selected for evaluation of phosphate solubilization by the colorimetric method. In the liquid media, the bacterial isolates showed phosphate solubilization ranging from 535.70 to 618.57 µg mL−1 in the NBRIP medium, and 374.20 to 544.28 µg mL−1 in the PVK medium, with the highest values produced by P. fluorescens. The best phosphate solubilization ability and higher reduction in broth pH, which indicates higher organic acid production, were achieved in NBRIP broth for most of the PSB. Strong correlations were observed between the average capability of PSB to solubilize phosphates and both the pH and total phosphorous content in the soil. The production of the hormone indole acetic acid (IAA), which can promote plant growth, was observed for all five PSB species. Among them, P. fluorescens obtained from the forest soil of northern Tunisia showed the highest production of IAA (50.4 ± 0.9 µg mL−1). -

Effect of Different Irrigation Solutions on the Diffusion of MTA Cement into the Root Canal Dentin
Publicaçãopor Martinho, José PedroOutros Autores: França, Sara; Paulo, Siri; Paula, Anabela Baptista; Coelho, Ana Sofia; Abrantes, Ana Margarida et al.(1) Aim: This study aims to analyze the in vitro infiltration of a silicate root canal sealer into dentinal tubules after using different endodontic irrigating solutions. (2) Methods: Twenty-nine teeth with single roots were separated into three groups according to the final irrigation protocol: G1 n = 10) = 17% EDTA (ethylenediaminetetraacetic acid) + 3.0% sodium hypochlorite (NaOCl), G2 (n = 10) = 17% EDTA + 2.0% chlorhexidine and G3 (Control group, n = 9) = 17% EDTA + saline solution. Root canals were filled using cold lateral compaction technique with MTA Fillapex sealer and gutta-percha. The sealer was labeled with rhodamine B. The teeth were segmented at the middle and third apical sections, which were visualized using 10× confocal laser microscopy to determine the sealer penetration percentage. (3) Results: In the apical section, no statistically significant differences were found between the groups regarding sealer penetration. In the middle section, Group 1 obtained the highest percentage, and Group 2 the lowest (p = 0.004). Group 1 also presented statistically significant differences in the Control Group (p = 0.031) and had close sealer penetration values. Meanwhile, the Control Group (p = 0.023) and Group 2 (p = 0.029) revealed a significant decrease of sealer penetration between the apical and middle sections. (4) Conclusion: The obtained results support that final irrigation with NaOCl promoted similar sealer penetration in the apical and middle sections. On the other hand, a significant decrease in the sealer penetration of the middle section was observed for the chlorhexidine and saline groups. Compared to other irrigant solutions, NaOCl promotes more uniform sealer penetration, which can correlate with better sealing and, consequently, higher endodontic treatment success. -

Apexificação associada a restauração corono-radicular em dentes anteriores definitivos: Série de casos
Publicaçãopor Dias, Sónia Isabel TeixeiraIntroduction: Treatment teeth with open apices is an endodontic and restorative challenge. The apexification technique has been associated with high success rates. In our eight cases the treatment option was apexification technique with MTA, tricalcium phosphate and BiodentineTM. MTA has been studied extensively and because of its excellent properties was the material most used in apexification technique in last years. Due to its inherent drawbacks other calcium silicate cements were used in apexification technique. The aim to treat this teeth is them long retention time. For that reason, the choice of a restorative treatment that reinforce or prevent root fractures like adhesive restoration technique extended into root canal system is essential.Aim: This case series aimed to evaluate long-term clinical and radiographic outcomes of seven nonvital immature teeth treated with apexification technique and adhesive corono-radicular restoration.Materials and Methods: This case series had the ethical approval of the Ethical Committee of the Faculty of Medicine of University of Coimbra. All five subjects were informed and signed the informed consent. A evaluation protocol was defined with inclusion of clinical and radiographic criteria. A blind observer evaluated each case following the established protocol. This case series included six subjects with eight teeth treated. Results: One of the teeth was lost because was considered a clinical and radiographic failure; The other six teeth were asymptomatic at clinical evaluation, but two teeth were considered a radiographic failure because of periapical radiolucency. On colour evaluation just one teeth had a grey shadow on the cervical area of the crown. Concerning to restoration evaluation, just one teeth have to substitute the coronal restoration.Discussion: Previous clinical studies reported a high success rate with apexification technique with MTA, and, less, with BiodentineTM to treat teeth with open apices. Many factors can influence the outcome of this technique but the combination of corono-radicular adhesive restorations and materials that have similar mechanical properties to dentin with this apexification technique may lead to a long-term success. In this case series, although with a limited number of cases but a long-term follow-up (range of five to twenty-two years) six teeth were considered a clinical success, which is in accordance with literature. Conclusions: Despite the limitations of our case series, the combination of these two techniques seems to be the best choice to achieve the long term survival of these group of teeth with necrotic pulps and immature root development. Key-Words: Apexification, Mineral Trioxide Aggregate, BiodentineTM, tricalcium phosphate, corono-radicular adhesive restoration, necrotic pulp, open apices. -

β-Tricalcium phosphate shaped by replica method
Publicaçãopor Porsani,N. K.Outros Autores: Trombini,V.; Ana,P. A.; Setz,L. F. G.Origem: OasisbrAbstract Biomaterials, such as tricalcium phosphate, can have different resorption rates in the organism that depend on their physicochemical properties, where the surface condition, such as morphology, size of particles and porosity, is important. In this work, we studied the coating of natural Luffa cylindrica matrix with β-tricalcium phosphate, using the dip replica forming method, in order to adjust the surface conditions to meet the demand of bone resorption. The materials were characterized in terms of specific surface area, particle shape, imaging in electronic scanning microscope, and zeta potential in water media. The suspensions used for coating of the matrices were produced with distilled water and sodium silicate as dispersant and carboxymethyl cellulose as a binder element. Rheological measurements were performed on rotational rheometer and viscometer. The products generated with 2.5 wt% of sodium silicate and 0.75 wt% of carboxymethyl cellulose, after sintering, had pore sizes consistent with high rates of reabsorption and porosity that suggest a future use as a biomaterial. -

Tricalcium phosphate sheets with chitosan obtained via aqueous tape casting
Publicaçãopor Freitas,A. K. C.Outros Autores: Silva,L. B. da; Costa,A. C. S. da; Peres,A. P. S.; Pergher,S. B. C.; Acchar,W.Origem: OasisbrAbstract Tricalcium phosphate (TCP) is a biomaterial produced in several biomedical applications, in addition to having biocompatibility and osteoconductive capacity. Biodegradable polymers of natural or synthetic origin such as chitosan and polyvinyl alcohol (PVA) have been used for tissue engineering applications because they have the properties of the native organic phase of bone tissue. Thus, the objective of this study was to obtain ceramic tapes based on TCP, chitosan, and PVA through the tape casting method. The viscosity data for all suspensions were ideal for the chosen processing. The thermogravimetric curves showed similar profiles, and it was possible to observe that a greater total mass loss occurred with the increase in the chitosan content. XRD reflections related to the β-TCP phase of the orthorhombic structure were found, showing no phase transformation. SEM analysis showed degrees of roughness on the film surfaces with little influence from the increase in chitosan content. -

Morphological, thermal and bioactivity evaluation of electrospun PCL/β-TCP fibers for tissue regeneration
Publicaçãopor Siqueira,Lilian deOutros Autores: Passador,Fábio Roberto; Lobo,Anderson Oliveira; Trichês,Eliandra de SousaOrigem: OasisbrAbstract Electrospinning is a simple and low-cost way to fabricate fibers. Among the various polymers used in electrospinning, polycaprolactone (PCL) stands out due to its excellent biodegradability and biocompatibility. However, PCL has some limitations such as low bioactivity, hydrophobic surface, and long in vivo degradation. Calcium phosphate ceramics have been recognized as an attractive biomaterial. They are bioactive and osteoinductive, and some are even quite biodegradable. Different contents of particles of beta-tricalcium phosphate (β-TCP) were incorporated in polymer matrix to form fibers of PCL/β-TCP composites by electrospinning for possible application in tissue regeneration. The presence of β-TCP particles promoted some changes in the thermal properties of the fibers. The immersion of PCL/β-TCP 8 wt-% fibers in simulated body fluid (SBF) caused the formation of a denser and homogeneous apatite layer on its surface. -

Effect of sterilization on the properties of CDHA-OCP-beta-TCP biomaterial
Publicaçãopor Morejón-Alonso,LoreleyOutros Autores: Carrodeguas,Raúl García; García-Menocal,José Ángel Delgado; Pérez,José Antonio Alonso; Manent,Salvador MartínezOrigem: OasisbrThe effect of the method of sterilization on the physical, chemical and mechanical properties of a new bone repairing material was studied. The material was obtained by thermal hydrolysis of beta-tricalcium phosphate/orthophosphoric acid cement and was composed of calcium deficient hydroxyapatite, octacalcium phosphate (OCP), and beta-tricalcium phosphate. Partial decomposition of the OCP was observed after sterilization for the three methods. Decomposition increased to the following sequence of sterilization methods: ethylene oxide; autoclaving; dry oven. On the other hand, mechanical strength decreased with regard to non sterilized material in the sterilization sequence: ethylene oxide; dry oven; autoclaving. The compressive strength was 8.5 ± 1.0; 9.0 ± 1.2; 8.2 ± 0.8 and 6.5 ± 1.0 MPa, whereas diametral tensile strength was 2.1 ± 0.3; 2.5 ± 0.1; 1.9 ± 0.9 and 1.6 ± 0.3 for the material sterilized by ethylene oxide, dry oven, and autoclaving, respectively. Several compositional and microstuctural changes were detected after dry heat and autoclave sterilization. Ethylene oxide sterilization had lesser effect on the chemical composition and strength than dry heat and autoclaving. -

Bio-C Repair - A New Bioceramic Material for Root Perforation Management: Two Case Reports
Publicaçãopor Toubes,Kênia Soares deOutros Autores: Tonelli,Stephanie Quadros; Girelli,Caroline Felipe Magalhães; Azevedo,Camila Grasielle de Sá; Thompson,Ana Carolina Tocafundo; Nunes,Eduardo et al.Origem: OasisbrAbstract These case reports aimed to describe the management of lateral perforation in the middle cervical third of the root in two maxillary incisors with pulp canal calcification using Bio-C Repair, with safe and viable clinical treatment strategies. Digital radiographic exams were obtained with different angles and analyzed using different filters. Cone-beam computed tomography (CBCT) images were requested to show the actual position of the canal, location of the perforation, and guide the strategic planning of the case. Subsequently, cavity access was prepared with the aid of dental operating microscopy. After perforation was identified, granulation tissue was removed and the original canal was identified and then dressed with calcium hydroxide. In the second visit, the perforation was filled with Bio-C Repair and the canal system filled with gutta-percha points and a root canal sealer (Bio-C Sealer). The teeth were restored with glass fiber post, 4 mm beyond the perforation level, and provisory crowns. Both teeth treated as described above were functional and asymptomatic with a 1-year clinical and radiographic assessment. The Bio-C Repair is suggested as a new cement option for the management of lateral canal perforations, with effective results as observed after a one-year follow-up. -

Si-tricalcium phosphate cement: preparation, characterization and bioactivity in SBF
Publicaçãopor Motisuke,MarianaOutros Autores: Carrodeguas,Raúl García; Zavaglia,Cecília Amélia de CarvalhoOrigem: OasisbrThere are evidences considering the effectiveness of Si on enhancing biological properties of calcium phosphates; however, there are not many works relating to the Si-alpha-TCP bone cement. The influence of silicon doping on the properties of Α-TCP cement was analyzed. Si-TCP was obtained by a solid state reaction employing CaCO3, CaHPO4 and CaSiO3 and powder was analyzed by XRD, FTIR, XRF and BET specific area. Cement samples were analyzed for their surface of fracture morphology, mechanical resistance and SBF bioactivity. Cement mechanical resistance was not satisfactory for biomedical application; nonetheless, sample's surface was coated by an apatite layer after immersion in SBF. Notwithstanding, to ensure that silicon is the element responsible for increasing the material's bioactivity it is necessary to evaluate the in vivo performance of the bone cement obtained in this work. -
