Caused by Alternaria alternata, Alternaria brown spot (ABS) is a major fungal disease of mandarins in Brazil, causing serious losses in commercial orchards of Ponkan tangerine (Citrus reticulata Blanco) and Murcott tangor [C. sinensis (L.) Osbeck × C. reticulata Blanco]. The response of 31 accessions/genotypes of mandarins and their hybrids to A. alternata, was evaluated in São Paulo state, Brazil. The incidence and severity of the disease in fruit were evaluated in the field, using diagrammatic scale, four times during 2008 and 2009. Based on these data, the area under the disease progress curve (AUDPC) was calculated. In parallel, in vivo and in vitro inoculations with A. alternata were made on leaves to confirm field results. Resistance and susceptibility was evaluated in controlled experiments by counting the number of lesions per leaf using a diagrammatic scale. Most of the genotypes were symptomatic after inoculation with A. alternata with disease most severe on the hybrids Nova, África do Sul, Ponkan and Murcott. Cultivars like Cravo, Nules and Ortanique that have excellent fruit quality and had lower AUDPCs, whereas Fremont and Thomas were highly resistant to the fungus and did not show any symptoms in the field or in controlled inoculations. The attractiveness and pronounced flavor of Fremont and Thomas fruit make them new options for the citriculture.

Compounds 1a-e, 2a, 3a, 4, 7-9, were tested for their antibacterial activity against Gram-positive and Gram-negative reference and multidrug-resistant strains isolated from the environment. Only 8 exhibited an in vitro growth inhibition of all Gram-positive bacteria whereas 9 showed growth inhibition of methicillin-resistant Staphyllococus aureus (MRSA). None of the compounds were active against Gram-negative bacteria tested. © 2018 MDPI. All Rights Reserved.

ABSTRACT: This study aimed to morphologically characterize the isolates of Ceratocystis fimbriata from yerba mate and to evaluate the effect of culture medium and temperature on mycelial growth and sporulation of C. fimbriata. For the morphological characterization of the 11 monosporic isolates of the fungus, slides were prepared to determine the dimensions of the sexual and asexual structures of the fungus. Four experiments were conducted to evaluate the mycelial growth and to evaluate the sporulation of C. fimbriata in different culture mediums and temperatures. The isolates of C. fimbriata from yerba mate showed perithecia with brown to black necks, divergent ostiolar hyphae, hatshaped hyaline ascospores, single-celled, cylindrical endoconidia, and globular to ovoid aleurioconidia. PDA and V8-agar media showed the highest mycelial growth. The average optimum temperature for mycelial growth and sporulation of isolates of C. fimbriata of yerba mate were 22.5 and 22.4 ºC, respectively.

Abstract We evaluated the effect of combined Rhizobium tropici, Trichoderma asperellum and plant growth-promoting rhizobacteria (PGPR) in beans crop. The hypothesis that strains of T. asperullum, R. tropici and PGPR combined could improve growth, biomass accumulation and beans yield was tested under greenhouse and field conditions. The treatments consisted of control, mineral nitrogen application and inoculation, isolated and associated with the following microorganisms: Rhizobium tropici, Bacillus subtilis, Trichoderma asperellum and Burkholderia sp. 10N6. Results were evaluated by shoot dry weight (SDW) and root dry weight (RDW), number of nodules and yield components. In greenhouse environment all the microorganisms behaved similarly, and the treatments inoculated with Burkholderia sp. 10N6 (IBu) and R. tropici (IR) stood out regarding the production components. In field conditions the treatments IR and IRTBa presented the highest values of SDW and RDW. Our results suggest that inoculation with R. tropici, T. asperellum and PGPR may promote beans growth and bring benefits to shoot and root accumulation, increase the number of nodules as well as improve yield components, contributing to a sustainable agriculture.

Fusarium oxysporum is usually found in foods such as corn, wheat, soybeans and barley, and can cause economic losses and risks to health in humans and animals due to the production of mycotoxins. The conventional microbiological test for fungal identification is based on morphological characteristics, requires specialists, and it is very time-consuming as it is necessary to carry out a culture of the fungus for two weeks before microorganism identification. Nowadays, researchers have made efforts to develop alternative analytical methods with advantages such as shorter time of analysis and better accuracy and reproducibility when compared to microbiological methods. In this way, in the present work, a qualitative analytical method to detect the secondary metabolites of Fusarium oxysporum in culture media by gas chromatography coupled to mass spectrometry (GC-MS) was developed with the intention of relating the presence of these chemical compounds to contamination by the fungus. The method comprised liquid-liquid or liquid-solid extractions with ethyl acetate from liquid and solid culture media, respectively, followed by GC-MS analysis, which was able to identify the presence of fusaric acid, 13-socosenamide, 5-(4-hydroxybutyl)-2-pyridinecarboxylic acid and toxin HT-2. The presence of these compounds was confirmed by high-performance liquid chromatography coupled to mass spectrometry quadrupole time of flight (HPLC-MS/QTOF).

In this work, an experimental design was used to analyze the influence of process parameters on the production of extracellular enzymes such as β-glucosidase and peroxidase, and their possible effect on the obtention of soluble and nanostructured silica from rice husk ash by the action of the fungus Fusarium oxysporum. Specifically, pH, fermentation time and glucose concentration in the culture medium were varied. Statistical analysis indicated that the silica synthesis in the aqueous medium was strongly dependent on pH and time. Although the glucose concentration does not exert a direct influence on the biosynthesis of silica, it is an important parameter in the production of extracellular enzymes. To prevent enzyme inhibition and provide higher dissolution of silica, it is recommended to work at a pH close to neutral with a glucose concentration of 3 g L-1 for at least 144 h.

The phytopatogenic fungus Lasiodiplodia theobromae, isolated from guava, was cultivated in rice for 32 days at room temperature. Extraction with CH2Cl2:MeOH (3:7), followed by chromatography fractionation of the extract provided ergosterol. From the fungus culture in Czapeck medium for 40 days at room temperature, were isolated isocoumarin cis-4-hydroxymeleine and an eremophilane-type sesquiterpene. The latter compound is being reported for the first time in the literature. Also, this is the first time that an eremophilane sesquiterpene is described for Lasiodiplodia genus.
