The aging of the worldwide population has associated an increased incidence and prevalence of several pathologies, such as cancer, skin lesions, and neurological disorders. To improve the therapeutic outcome, researchers have been involved in the development of new disruptive therapeutic products that provide personalized and more efficient healthcare solutions. Among them, inorganic nanoparticles such as those made of gold, silica, or graphene-based materials have been used by researchers and clinicians for cancer therapy. Inorganic nanoparticles present unique size- and shape-dependent physicochemical and optical properties that in conjugation with high loading capacities prompted their application as therapeutics, drug delivery vehicles, and imaging agents. Additionally, these structures can also be conjugated with targeting moieties or stealthing agents to further improve their accumulation in the tumor tissue and, consequently, enhance their therapeutic effect. Herein, the advances attained in the application of inorganic nanoparticles in tumor targeting, imaging, photothermal therapy, and delivery of bioactive molecules, such as drugs (e.g., doxorubicin), genetic material (e.g., DNA, siRNA), and immunotherapy mediators are discussed as well as its limitations and toxicity issues.

PEGylated graphene oxide (GO) nanomaterials have been showing promising results in cancer therapy, due to their drug loading and photothermal capacities. However, the recent reports regarding the immunogenicity of poly(ethylene glycol) based coatings highlight the importance of investigating alternative materials to functionalize GO. Herein, GO derivatives were functionalized for the first time with an amphiphilic polymer based on poly(2-ethyl-2-oxazoline) and were co-loaded with doxorubicin (DOX) and D-α-Tocopherol succinate (TOS) to be applied in chemo-phototherapy of breast cancer cells. The results revealed that POxylated GO displays the required properties for application in cancer therapy. Moreover, the screening of different DOX:TOS combination ratios showed that the 1:3 DOX:TOS molar ratio produces an optimal synergistic therapeutic effect towards breast cancer cells. Furthermore, this drug ratio had a lower impact on normal cells. POxylated GO was then loaded with this drug combination in order to assess its chemo-phototherapeutic potential. The delivery of DOX:TOS by POxylated GO to cancer cells induced a stronger therapeutic effect than that attained with the free drug combination. Furthermore, an even greater cytotoxicity towards cancer cells was achieved by exposing DOX:TOS loaded POxylated GO to near infrared radiation. Overall, POxylated GO is a promising drug delivery and phototherapeutic agent.

Graphene family nanomaterials’ (GFN) ability to interact with near-infrared light has propelled their application in cancer photothermal therapy. Furthermore, the graphitic lattice of GFN can adsorb different types of molecules, which has motivated their use in cancer drug delivery. However, the direct application of GFN in cancer therapy is severely hindered by their poor colloidal stability, sub-optimal safety, inefficient tumor uptake and non-selectivity towards cancer cells. To overcome these limitations, GFN have been functionalized with different types of materials. This review is focused on the different functionalizations used in the design of GFN aimed for application in cancer therapy, disclosing their role on surpassing the critical issues related to GFN-based therapies.

The incidence of fractures and bone-related diseases like osteoporosis has been increasing due to aging of the world’s population. Up to now, grafts and titanium implants have been the principal therapeutic approaches used for bone repair/regeneration. However, these types of treatment have several shortcomings, like limited availability, risk of donor-to-recipient infection and tissue morbidity. To overcome these handicaps, new 3D templates, capable of replicating the features of the native tissue, are currently being developed by researchers from the area of tissue engineering. These 3D constructs are able to provide a temporary matrix on which host cells can adhere, proliferate and differentiate. Herein, 3D cylindrical scaffolds were designed to mimic the natural architecture of hollow bones, and to allow nutrient exchange and bone neovascularization. 3D scaffolds were produced with tricalcium phosphate (TCP)/alginic acid (AA) using a Fab@home 3D printer. Furthermore, graphene oxide (GO) was incorporated into the structure of some scaffolds to further enhance their mechanical properties. The results revealed that the scaffolds incorporating GO displayed greater porosity, without impairing their mechanical properties. These scaffolds also presented a controlled swelling profile, enhanced biomineralization capacity and were able to increase the Alkaline Phosphatase (ALP) activity. Such characteristics make TCP/AA scaffolds functionalized with GO promising 3D constructs for bone tissue engineering applications.

    Publication
    by Campos, Adriano Rocha
    Origin: Oasisbr

    Publication
    by Gomes, Oto Leonardo Reis
    Origin: Oasisbr

Adaptation of marine organisms to environmental changes: native vs invasive species

    Publication
    by Ferreira, Maria João Figueiredo
Ria de Aveiro is a shallow coastal lagoon located in the northwest of Portugal that supports highly diverse communities of benthic invertebrates. Polychaetes are the most abundant group found, some of which are socio-economically important, as the case of the polychaete Hediste diversicolor. H. diversicolor is a native species in Ria de Aveiro and inhabits in soft-bottom communities of estuaries playing a crucial role in sedimentary processes and ecosystem functioning due to its unique traits. Recently, Arenicola marina was detected at Ria de Aveiro, being considered an invasive species in this ecosystem. A. marina is also a bioturbator and it is expected that its constant disturbance of the sediment surface can alter the infauna of the ecosystem. The production and applications of the nanomaterial graphene oxide (GO) has increased of in the last years, conducting to the release of this nanomaterial in the aquatic ecosystems, with consequences for benthic organisms, such as polychaetes. This study aims to assess the effects, isolated or in combination, of an abiotic stressor (GO nanosheets) and a biotic stressor (A. marina) on H. diversicolor. To achieve this, the study conducted several behavioral assays and biochemical markers. The obtained results demonstrated that exposure to different concentrations of GO had negative effects on burrowing activity of H. diversicolor. This species showed an unusual avoidance behavior, fleeing from lower contamination to higher ones. They were unable to escape from higher contamination compartments, but at the highest concentrations (1 and 10 mg GO/L) the bioturbation activity was significantly higher, which may indicate that H. diversicolor tended to move deeper in the sediment. Moreover, the presence of A. marina affected the behavior of H. diversicolor. Regarding A. marina, its bioturbation activity significantly decreased when exposed to GO, which could be related with the decrease of energy reserves, as protein content. Additionally, this species also showed an escape behavior from compartments with a higher GO concentration. Regarding oxidative stress, H. diversicolor seems to be more impacted, since LPO levels were higher in all GO concentrations and SOD activity significantly increased in lowest levels of GO. The obtained results indicate that A. marina may be able to some extent search for more prone areas for feeding, predation, reproduction, and migration under contaminant exposure. On the other hand, H. diversicolor spatial distribution may be severely constrained under both abiotic and biotic stress, while A. marina’s higher foraging activity may promote its propagation in the estuary. Concerning our results, behavioral tests, combined with biomarkers showed to be relevant tools and could be applied for the development of more environmental-realistic assessment and monitoring frameworks for estuaries.

Development of Au nanostars/graphene oxide paper for SERS

    Publication
    by Silva, Eduarda F.
Other Authors: Gonçalves, Gil; Fateixa, Sara
Graphene oxide (GO) has attracted significant attention due to its unique optical properties and tunable surface chemistry, making it an excellent platform for optical sensing applications. Combining GO with metallic nanoparticles allows for the fabrication of highly sensitive surface- enhanced Raman scattering (SERS) substrates for analytical purposes. Here, we report our research on chemical strategies to decorate GO paper with Au nanostars (AuNSs) for the SERS detection of methylene blue, a water pollutant model. Several preparative approaches were employed to evaluate their sensitivity to detect MB molecules, including polyelectrolytes, distinct graphene-based materials, and the deposition method of the AuNSs.

Interfacial assembly of zinc(II) phthalocyanines on graphene oxide (GO): stable "turn-off-on" nanoplatforms to detect G-quadruplexes (G4)

    Publication

Hypothesis The aggregation of phthalocyanines (Pcs) enfeebles their suitability as G-quadruplex (G4) ligands over time. It is hypothesized that the interfacial assembly of Pcs on graphene oxide (GO) influences intermolecular interactions, thereby affecting their physicochemical properties and inducing stabilization of Pcs in solution. Hence, the stacking of Pcs on GO could be tuned to create nanosystems with the ability to detect G4 for longer periods through a slow release of Pcs. Experiments Four cationic structurally-related zinc(II) phthalocyanines (ZnPc) were non-covalently assembled on GO by ultrasonic exfoliation. A comprehensive characterization of ZnPcs@GO was carried out by spectroscopic techniques and electron microscopy to understand the organization of ZnPcs on GO. The fluorescence of ZnPcs@GO was studied in the presence of G4 (T2G5T)4 and duplex ds26 through spectrofluorimetric titrations and monitored along time. Findings GO induced a re-organization of the ZnPcs mostly to J-aggregates and quenched their original fluorescence up to 98 % (“turn-off”). In general, ZnPcs@GO recovered their fluorescence (“turn-on”) after the titrations and showed affinity to G4 (KD up to 1.92 μM). This is the first report that highlights the contribution of GO interfaces to assemble ZnPcs and allow their slow and controlled release to detect G4 over longer periods.
