Recovery of free amino acids and muscle proteins from codfish (Gadus morhua L.) salting wastewater by sorption on Amberlite XAD16
Publicaçãopor Ferraro, VincenzaOutros Autores: Jorge, Ruben Ferreira; Cruz, Isabel B.; Castro, Paula M. L.; Pintado, Manuela E.BACKGROUND: The objective of this research was the valorization of an agro-food industry residue, namely codfish salting wastewater, through the recovery of valuable bioactive compounds. Approximately 200 L of salted wastewater are generated for each ton of fresh codfish, carrying protein and free amino acids at a concentration of c. 10 g L−1, and sodium chloride (NaCl) at c. 250 g L−1. RESULTS: Protein and free amino acids were selectively recovered by sorption with the Amberlite XAD16 resin. Food-grade acetone was used to elute amino acids while a solution of sodium hydroxide (NaOH) desorbed protein. Recovery of protein and of hydrophobic to neutral amino acids was 100%; NaCl was also partially recovered from the salting wastewater, at a level of 33%. Kinetics and mechanisms of adsorption and desorption were investigated. CONCLUSIONS: The process implemented can be considered as a valuable management option for codfish saltingwastewater, it can contribute to the fish industry sustainability and can serve as a source of nutrients. Free amino acids and protein extracts can find application in animal and human nutrition through incorporation into functional food and drinks, as well as in cosmetics and in pharmaceutical formulations. -

Solvent extraction of sodium chloride from codfish (Gadus morhua) salting processing wastewater
Publicaçãopor Ferraro, VincenzaOutros Autores: Cruz, Isabel B.; Jorge, Ruben Ferreira; Pintado, Manuela E.; Castro, Paula M. L.Codfish is worldwide mostly consumed salt-cured due to the highly appreciated sensory characteristics promoted by salt. During the salting process huge amounts of salted wastewater are generated – approximately 22% w/w of the codfish – carrying ca. 250 g/L of sodium chloride and ca. 10 g/L of organic compounds, namely proteins and free amino acids. In this study, the salt load of wastewater generated during the salting process of codfish (Gadus morhua) was successfully reduced by ethanol extraction. The effects of time, sample:ethanol ratio, pH and temperature on salt extraction by ethanol addition were assessed by a onevariable-at-a-time approach and then by performing a 32 fractional factorial design. The effects of pH were also assessed in absence of ethanol. The maximum amount of salt precipitated in the wastewater was ca. 33%, at a wastewater-ethanol ratio 1:1 (v/v), at a temperature of 0 °C and after 30 min. Proteins and free amino acids present in the wastewater significantly limited salt precipitation; in a blank solution (salt in water at ca. 250 g/L) a higher amount of salt, ca. 37%, was precipitated in the same conditions. Ethanol and temperature showed a linear effect on salt precipitation for both wastewater and blank solution however ethanol was the driving factor. Changes in pH did not result in salt precipitation either in absence or presence of ethanol. During salt extraction, no precipitation of free amino acids occurred in the wastewater while ca. 1.4% of protein fell into the precipitated phase along with the salt. -

Fish sarcoplasmic proteins as biomaterial for biomedical applications
Publicaçãopor Vieira, Sara Filipa FontouraIn 2014, fish captures and aquaculture supplied ca. 167 million tonnes of fish, of which about 87% were used for human food and 10% was destined for fishmeal and fish oil. The remain 3% is waste that can be used as raw material for direct feeding in aquaculture. Fish sarcoplasmic proteins (FSP) are soluble in water. Large quantities of those proteins are discarded as part of the waste water resulting from fish surimi preparation. FSP constitutes around 25-30% of the total fish muscle protein. FSP comprise heme proteins and enzymes, which are associated with the energy-producing metabolism of the muscle. Herein, FSP from codfish (Gadus morhua) were isolated, resulting in FSP extracts. Both FSP extracts and membranes were physicochemically characterized and their cytocompatibility with human lung fibroblasts (MRC-5 cell line) was also evaluated. By SDS-PAGE, it was possible to define the composition of FSP extracts. From the differential scanning calorimetry (DSC) thermograms, it was possible to define that FSP denature at 44.12 ± 2.34˚C. By circular dichroism (CD) spectroscopy, it was possible to defined that the secondary structure of FSP is mainly composed by α-helix structures. For concentrations lower than 10 mg/mL, no cytotoxicity was observed over 72h of culture. Further on, the FSP extracts were processed into FSP membranes by the spin coating. FSP membranes shown an uniform surface when analyzed by scanning electron microscopy. By CD spectrum, it was verified an increase in the amount of α-helix structures. The FSP membranes were more stable than the FSP extracts, since the denaturation temperature was higher as determined by DSC. FSP membranes were hydrophobic, with a surface zeta potential of -33.4 mV, showing distinctive mechanical properties, with a stiffness of 16.57 ± 3.95 MPa and a yield strength of 23.85 ± 5.97 MPa. Human fibroblasts cultured in direct contact with FSP membranes demonstrate their cytocompatibility until 48h. Based on these results, FSP can be considered a potential biomaterial recovered from the waste water of fish, constituting a pool of enzymes that has potential interest for wound healing applications. -

Evaluation of the potential of collagen from codfish skin as a biomaterial for biomedical applications
Publicaçãopor Carvalho, A. M.Outros Autores: Marques, A. P.; Silva, Tiago José Quinteiros Lopes Henriques; Reis, R. L.Collagen is one of the most widely used biomaterials, not only due its biocompatibility, biodegradability and weak antigenic potential, but also due to its role in the structure and function of tissues. Searching for alternative collagen sources, the aim of this study was to extract collagen from the skin of codfish, previously obtained as a by-product of fish industrial plants, and characterize it regarding its use as a biomaterial for biomedical application, according to American Society for Testing and Materials (ASTM) Guidelines. Collagen type I with a high degree of purity was obtained through acid-extraction, as confirmed by colorimetric assays, SDS-PAGE and amino acid composition. Thermal analysis revealed a denaturing temperature around 16 C. Moreover, collagen showed a concentration-dependent effect in metabolism and on cell adhesion of lung fibroblast MRC-5 cells. In conclusion, this study shows that collagen can be obtained from marine-origin sources, while preserving its bioactivity, supporting its use in biomedical applications. -

Cell-laden marine gelatin methacryloyl hydrogels enriched with ascorbic acid for corneal stroma regeneration
Publicaçãopor Alves, Ana L.Outros Autores: Carvalho, Ana C.; Machado, Inês; Diogo, Gabriela S.; Fernandes, Emanuel M.; Castro, Vânia Isabel Baptista et al.Corneal pathologies from infectious or noninfectious origin have a significant impact on the daily lives of millions of people worldwide. Despite the risk of organ rejection or infection, corneal transplantation is currently the only effective treatment. Finding safe and innovative strategies is the main goal of tissue-engineering-based approaches. In this study, the potential of gelatin methacryloyl (GelMA) hydrogels produced from marine-derived gelatin and loaded with ascorbic acid (as an enhancer of the biological activity of cells) was evaluated for corneal stromal applications. Marine GelMA was synthesized with a methacrylation degree of 75%, enabling effective photocrosslinking, and hydrogels with or without ascorbic acid were produced, encompassing human keratocytes. All the produced formulations exhibited excellent optical and swelling properties with easy handling as well as structural stability and adequate degradation rates that may allow proper extracellular matrix remodeling by corneal stromal cells. Formulations loaded with 0.5 mg/mL of ascorbic acid enhanced the biological performance of keratocytes and induced collagen production. These results suggest that, in addition to marine-derived gelatin being suitable for the synthesis of GelMA, the hydrogels produced are promising biomaterials for corneal regeneration applications. -
