Development of a bioactive membrane against vaginal infections based in a chitin-derivative

    Publicação
    por Conde, Alexandra Pereira
This work focuses on the valorization of residues, in this case, the crayfish by using it as raw material in the process of chitosan extraction. It also involves a study of the characteristics and properties of chitosan, to develop a membrane for the treatment of vaginal infections. The first step was the extraction of chitin from the crayfish shell. This was done using a chemical extraction method, characterized by successive reactions at different temperatures. The reactions consist in extracting the pigment astaxanthin with acetic acid, bleaching with NaClO, HCl solution for demineralization, and finally deproteinization with NaOH. The result was chitin, dried for 24 h at 50 ºC. This process had a yield of 8.42±3.35%. The extracted chitin was characterized with FTIR analysis and presents an acetylation degree of 97.6±0.2 %. Additionally, by acid hydrolysis, the percentage of pure chitin in female and male crayfish samples was determined. Females presented a higher percentage. Proceeded to the transformation of chitin into chitosan, and characterization. The deacetylation of chitosan was optimized, occurring NaOH solution in a ratio of 1:15 (m of chitin / v of NaOH solution), 5 h in a thermal bath at 100 ºC, and then 5 h at 130 ºC. The yield of this process was 63.4 ± 18.4 %, and the chitosan obtained presented a degree of deacetylation of 64.5 ± 0.2 %. A physical-chemical characterization using FTIR, DSC, XRD, and zeta-potential analysis was carried out. Bioactivities properties, antioxidant, and antimicrobial were evaluated. The microorganisms tested and for which it showed any antimicrobial capacity were gram-positive bacteria (methicillin-resistant Staphylococcus aureus and methicillin-sensitive Staphylococcus aureus), gram-negative (Escherichia coli), and fungi (Candida albicans). It was decided to develop an application in the field of biomedicine, exploring its potential as an antimicrobial agent. The final phase consisted of the production of chitosan membranes for the treatment of vaginal infections. It required the evaluation of their behavior, absorption, and degradation after immersion in a simulated vaginal fluid solution. The antimicrobial activity was also evaluated for typical pathogens of vaginal infections, and against Lactobacillus spp., common in the vaginal microbiota. The membranes produced with chitosan extracted from crayfish inhibited the growth of microorganisms. They also inhibited Lactobacillus spp., and, for this reason, the addition of probiotics/prebiotics in the formulation of the membranes is suggested. The membranes proved to be promising, being necessary adaptations, optimization of the production.

Seaweeds have long been used by populations on the coast of Portugal as fertilizers or food during periods of scarcity. Both uses have decreased dramatically since the introduction of chemical fertilizers and horticulture food. Even so, in the past decades, globalization has brought new interest to the consumption of these organisms, raising interest on algae as a healthy and tasty food. Much scientific research is underway on the properties of seaweeds, and it is proven that past uses are well-founded. S. Martinho do Porto, in the Centre of Portugal is the only location where the commercial harvesting of seaweeds still takes place. Gelidium corneum is harvested because this species produces high-quality agar, a much sought-after phycocolloid, which is used in the food industry as gelling or thickening agent. However, the species shows other properties that have been disregarded by the seaweeds’ industry. There are also some invasive species, namely Asparagopsis armata, which is causing considerable ecological problems to local ecosystems. Thus, the Mar2020 project Limo do Cais was approved to study the sustainable use the algal biomass from S. Martinho do Porto taking into account its industrial potential. In this way, the properties of G. corneum are being studied, to increase its commercial value. As to A. armata, it shows important antimicrobial properties that are being studied aiming the use of the algal biomass in the pharmaceutical industry. Thus, past uses of seaweeds will be discussed, as well as new potential uses that scientific research has uncovered.

Recent studies have dealt about the phenolic compounds and the nutraceutical properties of Sanguisorba minor Scop and about the possibility of their domestication to standardise the plant production. However, it is also known that the storage conditions can affect the bioactive compounds present in plants. Thus, wild (W) and domesticated (F1, F2, F3) S. minor samples were exposed to different drying methods (oven-drying at 60 °C until constant weight named OD or freeze-drying until constant weight named FD) and studied for their content in phenolic compounds, antioxidant, antimicrobial, cytotoxic and anti-inflammatory properties. In general, OD samples showed the highest nutraceutical properties and the highest content in phenolic compounds. The most abundant phenolic compounds in both drying methods were kaempferol-3-O-glucoside and caffeoyl ester, with some differences between wild and domesticated samples. Wild samples showed higher antimicrobial and cytotoxic activity than domesticated ones, except for the OD F3 and FD F2 samples that reported cytotoxicity against HeLa cells. This study provides important information to choose the most adequate methodology to retain phenolic compounds and nutraceutical properties of S. minor species. Further researches are necessary to standardise the domestication of the studied wild species and verify the highest efficiency of the OD method.

Food loss and residues have attracted the attention of many sectors around the world due to negative environmental and economic effects. Several studies identify this bioresidue with potential to be explored and used as natural ingredients, benefiting environment and economy, and producing functionalized products. This study aimed at the nutritional, chemical and bioactive evaluation of flours obtained from apples discarded as bioresidue due to their low caliber or non-standard format. The nutritional evaluation of the flour highlighted an interesting protein and ash content. Three free sugars were identified in the analyzed flour, with fructose being the main sugar and five organic acids, with malic acid being the most prevalent. Regarding fatty acids, monounsaturated fatty acids predominated, with oleic and linoleic acids being the major components. Seven phenolic compounds were identified by HPLC-DAD-ESI/MS, divided into three different families: phenolic acids, flavan-3-ols, and dihydrochalcone, with phenolic acids predominating, which may justify the antioxidant and antimicrobial potential evidenced by the flour. After characterization, flour was incorporated into two types of bread and the effect of this incorporation over the shelf life was evaluated. Overall, the nutritional and chemical composition of the apple flour obtained in this study makes it a viable alternative for application in the bakery and pastry industry, thus creating alternative products and health promoters for the consumer. Additionally, the reuse of apple bioresidue will allow the recovery of waste discarded annually without added value, in addition to contributing to a positive impact on the economy and the environment.
